Kit for Rapid Screening of HIV Protease Inhibitors for Discovery of Lead Compounds from Synthetic Chemical Libraries and Medicinal Plants


Abstract/Technology Overview

Human immunodeficiency virus (HIV)/ acquired immune deficiency (AIDS) syndrome is a critical infectious disease throughout the world. Long-term treatment with HIV drugs can cause the propagation of drug-resistant strains of HIV-1. Searching for a new treatment to overcome this problem is an active area of innovative research. The rapid assays are needed for a novel PI discovering to replace the current sophisticated techniques. Our team member at the Faculty of Associated Medical Sciences, Chiang Mai University has developed the rapid, simplified immunochromatographic strip test for detection HIV-1 PR activity and evaluation PIs, called "HIV protease inhibitors-based immunochromatographic strip" or "PIs-IC strip" [1-3]. Evaluation of PIs in clinical samples using the IC strip test revealed high relative accuracy, relative specificity, and relative sensitivity. Additionally, this strip can be applied for the detection of any HIV protease inhibitors (HIV PIs) in either synthetic clinical libraries or medicinal plant extracts. It can shorten the drug discovery processes without the requirement of expert scientists and expensive equipment.

Technology Features, Specifications and Advantages

To detect HIV protease (HIV-PR) activity and screen the inhibition of protease inhibitor (PIs) was developed by our research team. The novel immunochromatographic (IC) strip test was established by combining the proteolysis activity of HIV-PR and an IC reaction. Recently, we report this strip for evaluation of the biological activity of HIV-1 PR and efficacy assessment of this IC strip for semi-quantification of PIs in plasma samples from human immunodeficiency virus (HIV) -infected patients. We demonstrated the novel simplified IC strip test for semi-quantifying the level of PIs. This IC strip was feasible to investigate the PI concentration in clinical samples of HIV-infected patients with high sensitivity, specificity, and accuracy. In addition, the PIs- IC strip was comparable with the standard as HPLC. This strip could be applicable for facilitating the testing of the inhibitory effect of natural product or lead compound. Whereas, this strip was comparable with cell based-assay. The PIs-IC strip could provide as a high-throughput screening platform for discovery novel PIs from synthetic chemical compounds or natural lead compounds and for evaluating the biological activity of novel PIs to combat mutant HIV PR.
